Purification and properties of yeast transaldolase.

چکیده
Sedoheptulose-7-phosphate, originally identified as a product of photosynthesis in algae and higher plants (3), has been shown to arise by the action of transketolase on pentose phosphate (4, 5). This reaction occurs in a wide variety of animal (5, 6), plant (5, 7), and microbial cells (8-lo), and evidence has been obtained that the heptulose ester is an intermediate in the formation of hexose monophosphate from pentose phosphate (7, 11, 12, 13). With the isolation of sedoheptulose-7-phosphate from transketolase reaction mixtures (5), it became possible to study directly the conversion of this ester to hexose monophosphate. In an earlier publication from this laboratory (14) it was reported that this reaction requires a source of triose phosphate, and evidence was presented for the accompanying mechanism. The enzyme has been named transaldolase, since it catalyzes the

The transformation of pentose to hexoses was first observed by Dische (1) in hemolysates of red blood cells. Glucose 6-phosphate was shown to accumulate when ribose S-phosphate was incubated in a crude enzyme preparation from liver (2), and fructose 6-phosphate was identified by Clock (3) as the first product of the pentose-hexose transformation. It was shown by de la Haba and Racker (4) that t...

Background and Objectives: Laccase is the most abundant member of protein family that catalyzes the oxidation of substituted phenols. Laccases are used as biocatalysts for decolorization and bleaching in dye industries, detoxification in environment, and juice clarification in food industries. The present study aimed at producing recombinant laccase, purifying with high yield and fold, and char...
متن کاملPURIFICATION AND CHARACTERIZATION OF CELL WALLMANNOPRO TEINS OF CANDIDA ALB/CANS USING INTACT CELL METHOD
Virulence of the opportunistic yeast, Candida albieans, involves the interplay of many complex changes including the yeast-hyphae transition, which mainly involves protein changes. Cell wall mannoproteins are found to be the main cause of adherence of C. albieans to epithel ial cells in the first step of an infection process. In the present study, cell wall mannoproteins of intact yeast wer...
متن کاملExpression, purification and glycosylation analysis of chicken infectious bursal disease virus VP2 in yeast
Infectious bursal disease (IBD), a highly contagious and devastating disease in young chicken, is causedby infectious bursal disease virus (IBDV). To improve the immunogenicity of recombinant IBDV subunitvaccine, an attempt was made to find a new way to prepare IBD vaccine containing glycosylated mVP2antigen. Firstly, IBDV mVP2 gene (with a nucleic acid sequence encoding B cell epitope of IBDV(...
متن کاملProduction of Lavash Bread by Yeast – Salt Method and Determining the Rheological Properties of Its Dough
ABSTRACT: In this research, yeast-salt method was used to make flat bread dough. The yeast used for this process was Saccharomyces cerevisiae (PTCC-Code 5080), that was employed at the concentrations of 0.06% and 0.12% (w/w based on wheat flour). In order to carry out this research, a completely randomized design with three replications was used. Means were compared through Duncan's multiple ra...
